reaction was performed at 37C for 10 min. EDTA was added to contain a final
concentration of 20 mM in order to stop the reaction.

What you need is to prepare stock solution of each component of the reaction mixture and then to prepare the mixture by mixing the suitable volume for each component. For instance, let's say you have prepared 10x stock solution for each reagent then preparation of the mixture would consist in taking 1/10th of the final volume for each reagent and to top up to the final volume with water.
