There are some possibilities to measure H2O2 concentration. I want to measure extracellular hydrogen peroxide concentration by analyzing the culture medium via H2O2 dependent oxidation of Fe2+ in the presence of xylenol orange using the method described by Jiang et al. What is your advice?

sorry, I´ m not familar with H2O2 measuring by means of an reaction with Fe2+. So far I understand the problem, xylenol organe disturbs a colorimetric determination of H2O2. When xylenol orange exerts no fluorescence at an excitation wavelength needed for exitation of resorufine, you can try to apply the well established Amplex/horseradish peroxidase assay using the fluorinetric detemination of H2O2.

A brief comment: According to my experience, when H2O2 is added to cell culture media, it becomes rapidly decomposed. This caused by the medium content of minerals. So it might be frustating to try measuring of H2O2.

thank you for your answer. I am thinking about the Determination of H2O2 in culture medium with Amplex/HRP.
I know, that if you add H2O2 to your cell culture medium, that it has very short half life time. But to address this and ensure a nearly constant H2O2 concentration I try an Assay with Glucose/glucokinase +/- catalase. This is described by some authors... Let`s see how it works.
