I am looking for a quick fix because I am pressed for time. I know that you can add ampicillin directly onto LB plates, and was wondering if the same would apply to 3AT plates. I could not find any information online about this type of method. I had a mishap with the autoclave when I was trying to prepare the plates as I normally would. Thanks for your help in advance.

The purpose of adding supplement to molten agar prior pouring plates is to have a defined and even concentration in the plate at the end. If you spread supplement at the surface of an agar plate, the concentration will be uneven (higher on the surface and lower within the agar). In the case of 3AT, the actual concentration of 3AT is essential as it is selective for positive clones. In case of ampicilline the actual concentration in agar is less crucial because the selection is stringent enough on a wide range of concentrations.
