Can we use same amount of whole blood in place of serum/plasma for quantitation of hormones, while making sure RBCs are not lysed, in a non-chromatographic immunoassay? Would RBCs by themselves interfere? Also, would level of hormone be same in whole blood as in serum/plasma?

Serum testing of hormones has long been accepted by the conventional medical community as the standard for measuring hormones. Some kits uses plasma. RBC will interfere in the assay for measuring hormones, it's red colour of hemoglobin interferes in the assay.
I don't think RBC should be your major worry. Even if the assay isn't color based, there are all sort of other cells and stuff in there that can interfere. And to what extent I will have to know what assay and how it is detecting the target. And the same amount of whole blood (which I take it you mean volume) is NOT equal to the same amount of serum or plasma.
