In our lab, I found many old stocks of the trypsin-EDTA solution, which was manufactured 8 years before. it had been stored in 4°C for these many years. Is it advisable to use that for our cell culture purposes? Will it have any impact on the growth of cells (Mostly for Hela cells and mouse macrophage cell lines)?
Hi, I think that 8 year of storage are probably too much for every enzyme, also for trypsin that is a quite "highlander" enzyme.
I don't recomend to use it for your cells.
Try to let it more time reacting. I think it will interfere only on the time of its action
i don't recomend to use it.it is preferred to keep EDTA-trypsin just for 3 months.
As it is a very old enzyme it is not recommended to be used. But can you please do a pilot study using the same? Please covey the results if possible.
I would give it a try with some cells you can spare, even with different cells that you intent to use. Filtering would be and option to not carry over any posible germs. Look under the microscope every too minutes and see the cells are detaching. An old enzyme won't be harmful, just the activity will be dead.
thank you everyone. i tried that trypsin with mouse macrophage cell lines (RAW 264.7). within 2 minutes of incubation the cells have been datached succesfully and it does not affect the growth of cells too.. but i dont know how it will affect the other cells. so i dont want to risk my experiments. ill get the new product.
