Hi, I want to determine the enzyme activity of keratinase in the sample. I saw some literature regarding the same. where Keratin azure is mentioned as a substate for the same experiment. I do not have Keratin azure. kindly let me know if any other substrate can be used or not?

You could use colored, modified keratin-derivative as substrate like azo-keratin, which is a deep red orange compound. It is not commercially available as far as I know. You can prepare this substrate by coupling keratin (derived from various keratin sources) with a diazotized aryl amine to produce a chromophoric derivative, sulphanilic acid-azokeratin.
Keratin azure is a synthetic-colored keratin substrate which is available commercially and prepared from sheep wool.
There are also natural keratin-rich substrates available. See Table 2 & 3 of the attached paper.
