Hello!

We use homogenates of mouse brains for our aims (determination of cytokines by the ELISA method). After removing the brain, we freeze the tissue in liquid nitrogen, then store it for a while and then put it in a RIPA-buffer and homogenize in a Dounce homogenizer.

After preparation, we store the homogenate sample at -80. But we need to collect samples of homogenates for future research. Is it possible to store samples of homogenates if we essentially defrose them twice (first frozen tissue in nitrogen, and then thaw we defrose the homogenate)? Will we lose a lot of proteins during the defrosting process?
