is possible purify DNA from samples conserved in TCL buffer (qiagen)? We performed an experiment in which we decided to keep RNA for RNAseq but we observed a interesting phenomena that need to be analysed in DNA.
Best thing is to give a call to Qiagen support. They will certainly know. In my opinion, it is very well possible. Use DNA isolation kit on the lysed sample
Thank you everybody! I Asked Qiagen and they answer:
Dear Jimena.
Thank you for your mail to QIAGEN.
I assume, that you are referring to Buffer TCL from Turbocapture, right? Although we have not tested this, the best way would be to split the sample and do DNA extraction with one part and Turbocapture with the other part. However, if this is not possible:
1. You may try and use 200µl of the Buffer TCL Lysate as starting material in the QIAamp DNA Blood Mini procedure e.g. like blood. Again this should work in theory, but we never tested this.
2. Or you may try the Clean-up protocol of the Gentra Puregene Kit : Appendix C: Repurifying DNA Samples.
We hope we were of assistance to you. If you have any further questions, please do not hesitate to contact us again. Please also find our broad 24/7 online-support offerings at http://support.qiagen.com/
