I have two questions-
1. Can we knock out the mouse gene in both cell- and tissue- specific manner?
2. If so, can we further get the mice whose CD4+ cells in spleen only are lack of the gene? Is this impossible due to the migration of wild-type CD4+ cells into spleen? Or aforementioned migration will never happen though the T cells will migrate out of secondary lymphoid organs once differentiated sufficiently.

Cre/Lox system, as you said, can generate the mouse with a gene knocked out only in CD4+ cells.
But i am still worrying about if we can further specify the knock out in the CD4+ in a specific tissue, like only MLN CD4+ cells.
It will be great if you can please use your expertise again and give me a successful example of the aforementioned.

Tissue specific knockouts are achieved by placing the Cre gene downstream of a tissue-specific promoter. For example, TekCre will knockout floxed genes only in endothelial and myeloid lineages, AdiponectinCre will knockout only in adipose tissue, etc.

As you said, I find Jackson Lab do have CD4-Cre mice. But it seems impossible to confine the knock out in cd4+ cells of only MLN since we don't know any gene expressing exclusively in cd4 of the certain lymph nodes rather than the others
