Can we interchangeably use DPBS in place of Hank's Balanced Salt Solution (HBSS) for washing melanoma cells cultured in RPMI-1640 medium before trypsinization ? Will pH differences affect the cells during washing?
I used HBSS and also DPBS for 293T cells and did not notice any difference. The washing step is a short step and shouldn't influence the cells that much / at all (even if there would be a big difference between HBSS and PBS).
I routinely wash all my adherent cells in culture with PBS before trypsinization. The PBS wash is to remove any remaining serum in the flask, that will reduce the efficiancy of trypsin. The PBS was is just for a brief period which does not affect the cells unless your cells are highly sensitive to pH change. I used human and melonoma cells like B16F10, A375, CT-26. All the cell lines work well in cell based and xenograft models.
