Hello, I was wondering if transwell assays with permeable supports could be used to measure protein transfer between two different populations of cells. If I used a small pore size (less than 3 microns) and plated cells in the bottom chamber (instead of just media), and plated a different population of cells above the permeable support, would I be able to measure secretion of protein from the upper cell population and uptake of this protein from the bottom cell population (via immunocytochemistry or a western blot on whole cell extract?). Thanks so much for your reply! I’d like to measure progranulin, a secreted glycoprotein that is 88kDa in its fully glycosylated form. From previous work it seems to be discernable in media of cultured cells as well as in whole cell extract.
