i mean are chemical compounds of lysis buffer able to damage, probably.
lysis solution include: (2.5 M NaCl, 0.1 M EDTA, 10 mM Tris, 1% Triton X-100, 9% DMSO, pH 10)
no I havent control, i carried out comet assay on fish gill and liver tissue and used lysis buffer for all samples( treatment and control).
now, i want know that lysis buffer can making DNA damage and creat error in result?

are you doing comet assay on plant cells?
I agree with Kamran Shekh, however extended lysis of the cells may affect quality of agarose gels.

I also don't think the lysis buffer in itself can cause DNA damage.
However, it's always good to have controls (without treatment) to compare your results.
If the control has 0 to highest 10% DNA damage (background damage), then your buffer and procedure is fine.
The control(s) is the best judge of your procedure and solutions.
