I perform a plate count but there is no colony on the plate although they still normally proliferated in the MRSB culture. Please tell me why? Thank you so much.

I use MRSA for colony count. I dilute 10^7 10^8 10^9. After plating on agar media, I covered the plate by PE or Parafine and put them in Incubator. Everything is ok. I still don't understand what was happening ;((((. Thank you.

This isolation is from aquaculture (shrimp gut). I did but it doesn't work :((( thank you :).
Check the pH of the medium apart from inoculum, source and medium used.

I think pH is ok, Medium of agar plate and Erlenmeyer flask are same. When i inoculated on slant agar, they still grew normally. Thanks.

Yes, I did. I used to perform these experiment many times with some strains of LAB. I diluted from 10^0 to 10^9 :)))). nothing appeared on the agar plate, but I still saw LAB cells on microscope by using inoculum liquid. what's wrong with me? Thank you.
My LAB is Weissella cibaria.
Where are you putting your plates? Is it a 30 or a 37 degree incubator. Try incubating them in 30 degree and let me know. Also, reduce the dilution. And preferably, plate the crude suspension also. Don't wrap the plates with paraffin.
these bacteria require some growth factors ( fastidious), try to supplement some aminoacids in the broth to observe growth.
