we tried the Plasmid-Safe ATP-dependent DNAse in our lab to reduce chromososmal DNA contamination in DNA isolates after plasmid isolation. However, we could not see a hughe impact on our isolated DNA and finally sequenced the DNA without this treatment and the results were quite OK. So using this enzyme might not be necessary...

What kind of plasmids did you sequence? Are they the large conjugative plasmids? In my experience, the big plasmid extraction can be contaminated by chromosomal segments when perform plasmid sequencing in Illumina platform. Do you have any suggestions on removing the contaminants? Is the Plasmid-Safe ATP-dependent DNAse useful for this? Thank you.
