in our work we use this protocol:

one initial denaturation step at 95°C for 5 min followed by 30 cycles of denaturation at 94°C for 1 min, annealing at 50°C  for 1 min,  and extension at 65°C for 4 min

The PCR conditions are:

 one cycle at 95°C for 10 min, followed by 4 cycles of 5 min at 94°C, 5 min at 40°C and 5 min at 72°C and then followed by 30 cycles of 1 min at 94°C, 1 min at 55°C, and 2 min at 72°C, and the last extension at 72°C for 10 min.
