I do not believe that this would be possible. You have a very high ionic strength and mixed organic-aqueous solvent.  pK(s) are not known and ion-pairing definitely occurs.

The calculation is very easy (sum of mmoles of ions and non-ionis substances PER 1 L of solution).  However, it is rather pointless. Check the volume of water that you add in step (1). Otherwise, your osmolarity is not compatible with majority of animal cells. 

[1] Based on the components, I presume that you try to prepare intracellular-like solution for some sort of lysis or immuno-precipitation.  Correct?

[2]  Non-ionic detergent is likely to disturb or destroy integrity of the cell membrane (depending on final concentration).  If this is the case, osmolarity becomes a mute point, because it is critical only when permeability of cell membrane is intact.

Check all the volumes in your recipe, and ask your question again.  Make sure to mention what is the purpose of your solution.
