Hi, our lab has recently started working with primary hepatocytes in 3D form. The lot we are using has high basal activity which declines as the spheroids age. We are looking to undertake drug breakdown or metabolism with these spheroids - can I boost CYP expression by inducing with e.g. rifampicin and then adding the drug we want to be metabolized? Does CYP3A4 expression immediately halt once induction is stopped and if so should we then run the induction and drug breakdown experiment concurrently?
Any advice would be appreciated. Thanks.
