STZ is generally used to induce type 1 diabetes mellitus by intraperitoneal or intravenous injection at a dose of 45 to 65 mg/kg depending on the route of administration.
It seems that nicotinamide combined with STZ induces type 2 diabetes mellitus. If possible, I would like someone to enlighten me on this induction mechanism.
In case of T2DM, the main factor is to develop insulin resistance rather than hypoinsulenemia. If you have to develop moderate prediabetic stage then first try using HIGH FAT DIETS. As because STZ only destroy the beta cells creating a scarcity of insulin in the system thereby elevating the plasma glucose levels. But in case of T2D you have to create a state of systemic insulin resistance. So you should try HFD. You can follow my research article where we used this HFD regime to create hyperglycemic state.
