I would like to know how much formalin I should use based on the amount of tissue I have. When I complete HE staining my tissues look like they have shrunk. I am wondering if there is a protocol that could help me with the ratio of formalin to tissue. Thanks

The reason that I think it is the fixation step is I have tried multiple tissue processing steps and they all gave me the same result for HE staining. I have tried processing for long periods of time and short periods of time. Do you mind if I ask how long you dehydrate your tissue during tissue processing. I have also read that some labs rinse the formalin off before starting tissue processing. What are your thoughts on this? Thanks again!

Last but not least there is considerable doubt about (buffered) formaldehyde (alone) being the fixative of choice for such a task, remembering:
