Can someone confirm if this graph is correct? If there are any issues with it, could you please advise on how to optimize my MTT assay protocol?

When treating young HDF cells with a 100mM concentration of glucose, I observed the following graph. The cells exhibited rapid growth in the glucose medium, and those in the 100mM medium were too concentrated. I would like to confirm the appropriate high glucose concentration to be used.

I'm guessing that with the higher glucose concentrations, your cells are going hypoxic. You probably would have a difficult time keeping them with plenty of O2 in the medium.
