I need to perform southern hybridization of rhizobia nif genes so that I can determine their copy number in various isolates. Preferably I would like to detect with labeled antibody
You won't be able to detect with a labelled antibody, thats for proteins. A southern blot is for the detection of genomic DNA. Some discussions on this are here:
