There are many ways to measure cytokines secreted into medium by cells. Easiest way would be to look for a company that sells an ELISA-based kit for your cytokine of interest. Alternatively, you can purchase antibodies against your cytokines of interest and run Western blot on the media. If you are not looking at any cytokine specifically, or trying to figure out what cytokine is secreted, you can now buy arrays which allows you to identify multiple cytokines in one experiment, and will let you perform semi-quantitative measurement to determine if a particular cytokine level is different between different treatment-groups.

You are most welcome Erhan. In the past I have used cytokine arrays from Raybiotech and R&D Systems. However you still need to have an idea of what kind of cytokines you are looking for as they don't sell arrays that detect every cytokine. They have arrays grouped in catogories such as proinflammatory, angiogenesis. The arrays are also quite expensive so the idea would be to use the array to do an initial screen to find candidate cytokines then follow up using more traditional and cheaper assays such as ELISA kits.
I would also recommend ELISA. It's quite cheap (in the case of e.g. eBioscience Kits), easy to use and also easy to interprete achieved data. I also agree with Xiahui, that you need an idea which cytokines you want to measure.
I would recommend you first try the MILLEX multiplex cytokine array available with Millipore.They give you list of analytes(anti-inflammatory,inflammatory,angiogenic and survival factors) to be measured in the conditioned medium.They provide a high range standard curve ranging from 3.2-10,000 pg/ml and you only have to use 25 microlitres of the sample.
RAYBIO ARRAY is also a very competitive option.
Once you analyze,you will know the list of analytes of your interest and then you can go for ELISAs for various further experiments and analysis.
