I started working with Campylobacter and anaerobic organisms 35 years ago. I built all the microaerobic/anaerobic system to work with. It is very simple and you can try making it. Build a U manometer (see at the net). FIx it in a wall. Put behind it a scale from 0 (at atm ambient) to 30. Of course you have to buy an oxygen-free nitrogen and CO2 cylinders. Choose an appropriated solid medium, spread the sample over it (for anaerobic use two layers of solid media). Put inside the inoculated media, close the jar, adapt it with the vacum pump and the manometer and turn it on. Measuring the column of mercury, for example 27 cm up and down, you will have an atmosphere of 540 mmHg. Fill the anaerobic jar with nitrogen until 2,7 cm and the complete the atmosphere with CO2. It is enough to get an atmosphere of 5 % oxigen and 10% of CO2. To obtain an anaerobic atmosphere you have to wash three times the anaerobic jar with nitrogen, before you get the anaerobic athmosphere. I hope you understood. the first you should enrichment the bacterial by Bolton broth with the rate 1:9, incubate it under microaerophilic in 24-48h at 42oC, then cultivate bacteria onto mCCDA in 24-48h at 42oC. Extract DNA and make PCR with the specific primers...
