Hi guys. i am checking heat resistancy/susceptibility of my arabidopsis mutant. First i grow seeds on MS for 1 week and do heat treatment at 45 degree for 70 mins and allow it to recover for 3 days at 23 degree and then assess its survival rate after heat treatment. Arabidopsis seedling at this stage show 2 cotyledons and 2 true leaves. Plant that show total all whitish leaves is consider as dead. But how about plant that show only single leaf whitish or 2 whitish leaf and remaining two is still green? Do they consider alive or dead in survival rate assay?Tq!

first of all, it is very tricky to do heat stress on plates.
e.g. I am not sure from your description, whether or not your plates were put into darkness. If not, you will probably measure photo(respiration) damage, instead of heat (see any of the articles by Schöffl F.).
To come back to your answer:
You said, you measure survival rate. Well, if some leaves turn white, but your plant recovers, then it survives!
Either you measure 'white leaf area' or any other quantitative measure (e.g. total Chlorophyl/weight) - BUT, it is rather speculativ to assess 'survival rate' after 3 days. If you'ld count number of plants that survived until phase shift to bolting or seedset, I can accept that those had survived.
Thank you Dr Wanke for your suggestion. I did the treatment in closed chamber so it is in darkess. As i referring publication of FCA thermotolerance by Sangmin Lee (New Phytologist (2015), they assay the survival rate of heat treatment after 3 days recovery. I could not see any definite survivial/dead based on their publication figure. So i think i should let them recover more to see if the plant survive as you suggested.
