I tried this reaction starting from FMOC protected Asp with serine (OtBu protected) in presence of DCC/HOBt and then next step deprotection with piperidine and so on...But this strategy does not seem to work so well.
Have you tried HBTU and oxyma? Warming the reaction mixture in DMF upto 60-80C greatly improved coupling as compared to RT. Or try it manually on solid phase resins which will improve the synthesis and coupling over solution phase. You can also try PyBOP as coupling reagent which is used for difficult coupling steps. Also take care of the solvents that should be dry!

Thank you for this suggestion. I think I can try this reaction in DMF at 60-80 degress. We do not want to use solid phase peptide synthesis, though I think it may give better results but right now we do not have reagents/chemicals for that.
I would suggest to go with either Z-protected or Boc-protected amino acids, DIC/ Hobt should work efficiently. All the best!
