We would like to confirm mRNA data on up-regualtion of BMF by Western blotting. However, most of the publications suggest that commercial antibodies do not really work. I was thinking about Sigma Aldrich Atlas Antibody - however, this was only verified with overexpressed recombinant protein. Any suggestions? Thanks.
Did Sigma say that they tried on endogenous BMF but it did not work ? Or they just did not try ?
I will just try on your endogenous BMF. If you load enough material and have an efficient
transfer, you should be able to see your band using very sensitive films and hyper ECL.
Once/if you get a signal:
For the specificity of the signal, I will ask them if they can sell you the peptide used for the immunization to produce the antibody. I will do the incubation with the primary with or without the peptide in the incubation bath.
There are plenty of suppliers that claim their Abs will blot BMF. Some of them offer trial sizes (e.g. http://www.biovision.com/documentation/datasheets/3334R.pdf) or guarantees for specific applications (e.g. http://www.ptglab.com/Products/BMF-Antibody-18298-1-AP.htm) which may make them worth a try.
