Can reserpine be washed off?

Hi guys,

I am using Hoechst 33342 for sorting live cells and using reserpine to improve Hoechst staining (I have observed it improves the staining and there is a logical explanation to that). My only question is can we fully wash off reserpine after sorting as it is reported to have bad effects on cells? Have anybody used it before ??

Hi Ankur,

Yes we have used it for Hoechst staining. It is a pump inhibitor to stop the dye being eluted from the cell. Yes it can be washed out. If anything, depending on the cell type we had more an issue if we didn't keep adding it. We use to use Verapamil in addition to ensure the dye stays where it is meant to.
