I have samples which I need to extract for analysis such as RNA, protein and cellulose test.
I will first grind the sample to powder using liquid nitrogen for RNA extraction, then the rest of the sample will be used for cellulase/protein extraction and cellulose test. For cellulose test I will freeze dry the sample but for protein extraction, is it ok if I store the powder sample in -80C until extraction or can I freeze dry it before extraction? (I have multiple samples, so I cant do everything in one day. )
Freezing destroys cellular structure and components. Using freshly cut plant material is what makes my patent work. (See Systems and Processes for Producing Biofuels from Biomass; US8641910B2.) Protein does change as a result of freezing, but evidence can be minimized. (See Lavinia M Lewis, et al. study "Characterizing the Freeze-Drying Behavior of Model Protein Formulations.")
