In polymerase chain reaction, we obtain amplified DNA segments that can be transcripted to produce RNA. According to this method, we might be able to achieve RNA directly from DNA in this reaction.
A T7 RNA polymerase promoter is needed for the artificial transcription so your PCR product (dsDNA) must include this promoter sequence at 5' site (you have to design your forward primer adding this T7 sequence) and reverse primer should include stop codons.
After PCR with these primers, you can use T7 RNA Synthesis Kits to produce your RNA from your PCR product.
