I found an interesting article about MSCs migration which is guided by FBS concentrations (http://onlinelibrary.wiley.com/doi/10.1002/jor.20668/pdf). My questions are:
1) How cells sense the food sources?
2) Is FBS a good chemoattractant universally (i.e. regardless species)?
3) What components in FBS make it a promising chemoattractant?
4) Seems to me MSCs can sense PDGF better than FBS. What is the reason for that?
Hi Long, absolutely yes. FBS is a good chemoattractant. By using commercial inserts with filters for the migration assay, and by positioning below mediums with different FBS concentrations (1% and 10%), we observed an increased migration toward the 10% FBS-supplemented medium.

Hi Filippo, since FBS has a complex composition, what contained in FBS do you think related to the chemotaxis of MSCs?
Also, seems to me you use the Boyden chamber for the experiment. Do you see the cells really move toward higher FBS concentration? Also, how do you know for sure that cells migrate but not proliferate? Seems to me FBS also improves the proliferation so making the number of cells higher.
Hi Long, I don't know which is the specific component, that was not the aim of my experiment. FBS is crucial for cell proliferation; for this reason, you have to inhibit the proliferation before performing the migration assay.
