I wanted to ask if anyone ever managed to create a regular shaped (for drug testing evaluation) 3D spheroid for MDA-MB-231 on agarose coated 96-well plate without any addition of methyl-cellulose, Matrigel etc. I am NOT going to use polyHEMA or any ULA plate, or round-bottom plate, or V-shaped bottom etc. Probably, I am going to use either hanging drop, then transfer the spheroid (if it formed) into the 96-well agarose coated plate or the liquid overlay method with centrifugation of the plate on swing bucket before incubation. So, is this method plausible for MDA-MB-231 (it works for some other cell lines)? Or is there any specific protocols for it? Thank you.
I tried several different methods to make MB231 spheroids, and 3D Bioprinting was the best one. I could not make nice spheroids in ULA plates, in hanging drops, in U-shaped wells, etc. Also, it is easier to make spheroids from this cell line when you combine MB231 cells with fibroblasts.
Like Vilma, we also tried to cultivate MDA-MB231 cells as spheroids in different culture conditions (agarose-coated surfaces; non-attachment 96-wells) - without getting the classical spheres. Having had this experience also with other dedifferentiated carcinoma cell lines, which also formed rather amorphous but tight aggregates, this is most likely a characteristic feature of these cells.
