I have worked with other BSL2 bacteria before (F. nucleatum) and we used an open flame to pick colonies and inoculate cultures. If I am working with staph aureus--i.e., picking colonies to put into a solution for downstream analysis--is working with an open flame sufficient, or is it necessary to work in a biosafety cabinet?
It depends on the strains of Staphylococcus aureus that you work, for example, if they come from clinical samples you can have very virulent strains and for your safety and that of your laboratory works in laminar flow hood with the highest possible biosecurity. If the strains are from a laboratory collection or are ATCC, check their characteristics, some isolates could be worked with the use of a lighter and a minimum sterility area.
