1) I would like to do a whole-cell e-phys experiment where I wash in an activating antibody against beta-1 integrin, but would the presence of this antibody then affect the ability to form effective seals in the same dish?

In relation with first question. No the antibody does not interfere with patch clamp.
So, you want to wash in your antibody before or after you form a seal? I would form a seal first in BL conditions, record my signal (whatever it is) in Control and then wash in the antibody. Now, if you need to pre-incubate your cells with the antibody first (because of whatever reason), then just try and see! In theory, It shouldn't affect your ability to get seals but who knows what your compound does to the cell's general state of health (cell volume, Ca2+ leakage...etc.).Try it and if it does not work, Go to plan A (i.e., patch in BL solution and wash in the ab later).
