I am new to patch clamping and I am trying to see inward voltage-gated current in SKOV-3 cells grown overnight w/o serum. Solutions are (mM) bath: 130 NaCl, 5 KCl, 10 BaCl2, 10 HEPES and pipette: 60 CsCl, 60 CsOH, 60 aspartate, 2 MgCl2, 10 HEPES, 10 EGTA. I go whole cell and get seals of about 5 Gohm and hold at -80 mV. I record a single sweep applying a step to -10 mV (at 65 ms) for 50 ms with P/4 subtraction beforehand.

These look like they could be capacitance transients or some other noise in your recording set up to me. The stereotyped magnitude and brevity of the responses seem to be clues.
Capacitance transients? Neither Na+ nor Ba++ inward currents were found. This cell (SKOV-3) might not be electrically excitable.
Thanks for your responses I think they are capacitive transients as I was unable to adequately compensate for these using the model cell
Assuming that these results commonly occur, SKOV-3 might not display voltage-gated inward currents. BaCl2 at a concentration should induce large inward current with the slowed current inactivation (barium inward current).
