This strongly depends on the detection antibodies used in the rat ELISA KIT and its specificity. This information should be given in the manual of the ELISA (see online PDF).
Although antibodies may cross-react from rat to human IL-10, usually the cross-reactivity is not sufficient to make a rat cytokine ELISA reliable for measuring human cytokines. Human IL-10 ELISA are widely available and so it is difficult to justify using a rat ELISA.
Iain Martin Sheldon Sir, thanks for feedback. While checking the kit in its seller's website, I read some peer reviewed articles in which the Rat ELISA kit was used. But, I found the authors using the human umbilical cell culture supernatant. So, I was confused on the reactivity of IL-10 of human and rat.
