Totally wrong. Dissolve plant extract  in pure DMSO. Say, make a stock of 100 mg plant extract in 1 mL of DMSO. Then make dilutions in 5 % DMSO (5 mL DMSO added to 95 mL of H2O) to get 10 mg/ ml solution by taking 1 mL of stock solution above and adding it to 9 ml of of 5 % DMSO , and then 1 mg/mL solution, then 0.1 mg/mL and so on. Try all concentrations for discs against some bacteria and see where you get zones of inhibition. Prepare negative controls the same way without the plant extracts. Serial dilutions! If you get turbidity, then slow centrifuge to get the particulate turbidity-inducing useless particles/ compounds. Thanks,

Sure- please use "pure" DMSO to get it dissolved at first, and then make serial dilutions in 5% DMSO in water or less as more than that would be "toxic" and antimicrobial too. Also, use the same dilution of pure DMSO as a "negative control" on same bacteria under same conditions to "minus" he effect of that

Yes you can use DMSO to dissolve the extract for antimicrobial study. 

For Test= Extract dissolved in DMSO ( in various concentrations)

Control = DMSO

DMSO is toxic. Take that too into your consideration!

thanks to all contributors, I mean by control is to perform the antimicrobial activity of DMSO alone as control....maybe is better to perform control with mixture of DMSO 5%: H2O 95 %.

I dissolved the plants extracts in 5 % DMSO then I added 95% water, but I got turbid and creamy stock solution, can I proceed by applying this turbid solution on paper discs on agar??

Yes you can dessolve in DMSO. if you need further confirmation , you can use DMSO as a control.

You need to try it out. But for heaven's sake do not inject this DMSO extract soln, in either GC, HPLC or even MS. You may have to pay a heavy price if you do so.

Yes, you can use DMSO for the antibacterial activity because there is no activity against the microorganisms especially bacteria and fungus as in my all previous antimicrobial experiments. You can also used Dimethyl Sulfooxide as a negative control against the microbes and used any antibiotics as a positive control.

Ya sir,  exactly you can use DMSO as a diluent and it is universally accepted. Sometimes instead of DMSO you can use DMF (Dimethyl Foramide) also and there is no much difference between them. One more thing while selecting this diluent, make it sure dissociation capacity of concern extract with DMSO or DMF, because if crude extract concentration exceeds there will be problem. Simultaneously you can keep DMSO or DMF control disc also.

DMSO is standard solvent to study antimicrobial assay. DMF in some case use, but better is DMSO. I also done a same type of experiments, u can read it if u wish might be helpful for u!

You could use any solvent based on two criteria: 1. The solvent should completely dissolve your extract at the maximum concentration, you want to test its activity 2. The solvent should not have antibacterial activity.  

we have tried the DMSO on GRAM positive and negative and yeasts with the rate of 10 micro-liter per disc and it has no effect on the growth.

You can use DMSO to dissove your methanolic extract because it is a universa solvent and have no or very little antibacterial activity. u can use it as a control when performing your antibacterial activity test. Good luck

You have to optimize the dose of DMSO with reference to dissolve plant extract. You have to determine how much dose of plant extract is able to dissolve in minimum concentration of DMSO. Because DMSO become toxic in a dose dependent manner.

You can use normal saline as control. The dose of DMSO optimized for plant extract will be used as a treatment to investigate the effect of that dose of DMSO

You need to dissolve maximum plant extract in minimum conc of DMSO, because DMSO is toxic at higher conc.
