Actually, immediately after the delivery of Opti-mem (Gibco), it was accidentally kept at -20Deg C. It was not opened. So if i use it now for transfection media preparation, would it affect that much? Or should i purchase a new one.....
Depends on whether you have passed the shelf life given on the bottle. If no, then it should be alright. If yes, then it is suspect. If you want to store medium or serum or supplements, the best temperature is -80C (aliquot serum, and supplements to amount you will need, repeated thaw-freeze will degrade the material). At that temperature the products go into a suspended animation state and only continue their shelf life degradation if placed at higher temperatures, such as -20C, ambient, or 37C. We have problems such as you describe early on and for the past 25 years have stored our reagents at -80C to extend their respective shelf lives.
