Hi, I am working on ELISPOT assay and I don't have any luminometer and ELISPOT reader.
In this case, can I use ELISA reader (Biorad 680) instead? (Details about reader is described below)
I have no idea what ELISA reader, luminometer and ELISPOT reader are different.
I am using BrdU Kit and ELISPOT Kit which are both colorimetric.
1. Wavelength range : 400 - 750 nm
2. OD range : 0.000 - 3.500 OD
3. Resolution : 0.001 OD
4. Accuracy : ±1.0% or 0.01 from 0.000 to 3.000 OD at 490 nm
5. Reproducibility : 1.0%, or 0.005 OD from 0.000 to 2.000 OD,

I think that depends entirely on the type of kit you have for BrdU. Whichever antibody has the enzyme tag, that will tell you the type of detection system you will need. The ELISpot data from an actual ELISpot reader will give you data on the number of instances of cell proliferation due to incorporation of thymidine into the duplicated DNA. A standard plate reader can give you data analogous to that, but only if the secondary antibody (or whichever has the enzyme label) generates a signal via substrate which can be detected (i.e. a suitable wavelength). And the luminometer the same, but only if the enzyme label is capable of producing that signal, and you have the proper substrate, of course
