I just wonder that we can use CAS assay to determine the siderophore in culture media that contain some amount of iron or not ? According to the principle of CAS assay, the siderophore binds the ferric iron, releasing the free dye, which will change the color of the dry from blue to orange. If in the media is already have iron, siderophore can bind with iron in the media instead of iron that form complex with dye. So, can we still use this assay to measure the siderophore production in iron containing media?
