I need to purify platelet from human blood. The ACD tube available from BD only come in glass. May I know if it is ok to use it to collect blood for platelet isolation? I want resting platelets.
Yes, of course, you can use ACD glass tube to collect blood for Platelet isolation. But you have to isolate platelet immediately (within 2 hrs), because they might get activated easily if you keep for a long time. (if you want non-activated platelet)
Yes, you can definitely follow Palikhe's suggestion but siliconized tubes are generally recommended for isolation of platelets as they are adherent cells and probably the ACD tubes from BD might be siliconized.But for isolating resting platelets time is one of the major factor indeed.
It depends on what do you intend to do with isolated platelets. If you need non-activated platelets, I would suggest you to do it the way that I do platelet separation for more than 20 years: I use sterile plastic (50 ml polypropilene) conical tubes with cap produced by BD, Falcon, Cat. No 352098. I need non-damaged, non-activated cells that will be labeled with In-111 oxinate, and thereafter re-injected to the patient in order to discover the mechanism of thrombocytopenia. You will find the method that I use (in text and as a ppt presentation, together with referenced) enclosed. On the other hand, if the quality of separated platelets (their activation or aggregation) does not matter to you, you could use glass tubes.
Thank you for all the valuable feedback. I am planning to do omics analysis on the isolated platelets. How do you usually store the platelet? As pellets or aqeuous? for how long at -80oC?
I also use the BD ACD tubes for collecting blood for washed platelets. If we plan to keep the platelets for other studies we usually pellet them and then freeze the pellet immediately at -80 C. Not sure how long they keep for and this will only work if you are planning on lysing the platelets before analysing otherwise you may want to at RIPA buffer or something similar.
