I did a plasmid transformation using BL21-DE3 bacteria yesterday for protein expression, however I couldn't grow them in LB, so I wonder if I can store the BL21-DE3 at 4℃ for one day, or I need to do a transformation again. Thank you!
Yes you can keep them in 4C for several days. Although if you have a lot of colonies, or large-sized colonies, you may loose the opportunity for picking up single colonies.
You can store the plates at 4C. However, there are chances that some false transformant may occur or you may face difficulties to pick single colony/transformant if there are many transformants/colonies on your plates. So it’s always better to use the fresh plates. I hope it will help you.
Definitely. I did that on occasions when I was unable to pick the colonies for screening. Storing the transformed plate for a few days are fine. I sometime store my plates with transformed colonies from glycerol stock at 4℃ for a few weeks. I'll usually discard them after a month as the bacterial viability will start to decrease. But of course, the best practice is to pick and screen the colonies not long after transformation and make glycerol stock for the positives, in this way, it saves you the hassle of having to do transformation everytime for your experiment.
