I encapsulated Collagenase in Chitosan Nanoparticles and I would like to sterilize it before invivo testing , however i have a very smll volume that might be lost if i use 0,22 um filters so i was looking for alternative methods of steralization. I also modify the surface of my particles with peptides and im worried that UV might affect it.
Hi Sara, sterilization by UV is very inefficient because UV does not penetrate very far. I think the easiest way is to add antibiotics and antifungals to the NPs before in vivo experiments.
Yes, you may sterilize protein using UV lamp. But it depends on protein type and intensity (W/m2). Protein absorb UV between 280 and 200 nm.

Can you please explain what do you mean it will affect chitosan or collagen. I care that my chitosan nanoparticles to remain intact after sterilization to be able to inject it IV.
Hi Sara, bottom line:
-UV sterilization is inefficient and may cause crosslinking of the NP proteins.
-0.22 micron filtration may result in loss of NPs.
-Autoclave is not an option.
So your options are limited.
My recommendations is that you test your NPs for endotoxin. If they are high in endotoxin, run them through a detox gel then incubate them with antimicrobials and inject them, Controls are samples treated the same way but without NPs.
