We are planning to use Human XL Cytokine Base kit for evaluation of concentration of several cytokines. The plan was to measure concentration of cytokines both in blood serum, and cell culture supernatants. But in the description of the assays it is stated that reconstitution of standard solutions depends on the nature of the samples: The RD6-65 calibrator diluent should be diluted twice for supernatants. Expected control values differ for cell culture and serum, but not significantly. E.g.: IL-10 612pg/ml for serum, 627 pg/ml for cell culture (Low control); 5383 pg/ml for serum, 5273 pg/ml for cell culture (High control). Will it be still acceptable to measure different types of samples with the same plate?

Does anyone have any experience with "stretching" R&D/Luminex Multiplex kit for larger number of samples? We did that with Merck kit, proportionally reducing the volume of reagents to be added, and thus measuring up to 100 samples with one kit using 2 plates.
