I am using bouin solution for fixation of cryosection tissues: i would fix my tissues in bouin solution at 58C. I plan to do Immunofluorescence on these tissue sections. I am wondering if reused heated bouin solution might affect the fixation step?
If you plan to do IF staining, probably you have to change your fixative depending on the type of antigen and antibody. Some IF staining do not work well after bouin fixation.
Hi, Bouin's is the best fixative for histology - paraffin embedding and H&E staining. For cryosections, formalin would be better fixative. If you are planning for immunofluorescence you may look for appropriate fixation to retain antigenic ability of tissue/cells. Fixation at
