Dear all,
I would like to revive and culture L. acidophilus from the purchased L-dried sample. As I do not have anaerobic facilities, I am thinking to grow them in aerobic conditions. Could someone suggest if it works. I will be glad if anybody will help me in getting an authentic protocol for such growth studies.

Did you receive any growth recommendations with your culture? Most lactobacilli tolerate oxygen but some strains may grow poorly in the presence of oxygen. You may tray growing your strain in MRS supplemented with 0.5% cysteine if you think that oxygen may be an issue for your strain. Using screw capped tubes or agar plates sealed with parafilm is also recommended.

Thank you for your suggestions. The organization recommended MRS medium for revival and cultivation. However, no instructions are there for anaerobic cultivation. In addition, I would like to know if these strains can grow in LB medium. Are MRS medium mandate for their growth ? I will be grateful if you will share any experiences in growing L. acidophilus in LB just like E.coli.

Dear Bikash,
Lactobacilli do not grow in LB. They are auxotrophic for a number of amino acids and vitamins and require a sugar as a carbon source so that you need a complex medium that provide all these required nutrients. MRS is the "standard" medium for growing lactobacilli and it is commercially available.

From my experience with lactic acid bacteria in general, it is very possible. The issue is in aerobic conditions, it takes much longer to grow. Sealing MRS agar plates with parafilm or growing in screw capped bottles is also a viable option in absence of main anaerobic culture set up.
Hi,
Inoculate your strain in screw capped tube contained MRS broth then put sterile vegetable oil to maintain anaerobe condition. Incubate at 37 oC.
I am working on the same task now. Did you figure it out? LAB supposed to be air tolerant, but I am sure there are some strains that are not.
