According to the vendor, vesicle samples isolated (from culture media) using this kit can be used for proteomic analysis. I have had difficulty in getting a sample that can be used for this. I have followed the kit partially, stopping after the centrifugation and supernatant removal post TC incubation, then resuspending in PBS. This caused issues because the polymer precipitation material was still very present in the sample and clogged up the filter. While it caused technical issues, the LC/MS was successful in terms of the results. I then followed the kit using all the steps (further purification), but then my sample was resuspended in a large volume (200-300uL) in unspecified buffer. To try and reduce the volume and remove the buffer, an acetone precipitation was completed. This yielded no results after LC/MS. Has anyone had success using this kit to isolate a sample for LC/MS?
