I'm conducting experiments using carboxyl-amine coupling via high concentrated EDC/NHS reagents in aqueous condition(MES buffer & PBS). After few minutes, some bubbles(gas?) were generated in the bottle. I were trying to check which elements can produce bubbles, but I couldn't find the solution. Could you tell me why there were some bubbles during EDC/NHS coupling?

Hi Seung Woo Choi . The EDC/NHS reaction by itself shouldnt produce any bubbles. You could degas the buffer before adding the EDC/NHS.

Stephanie K Seidlits Thank you for your comment! The paper you recommended said "EDC and NHS generated CO2 bubbles during the process of activating the carboxyl group of the HA", so I will check about this.
I am having the same problem. It looks like the mixture EDC/NHS in aqueous solution forms bubbles using a microfluidic pump. Probably due to a high concentration.
In the paper that I share herein, it is reported that trace amounts of ethyl isocyanate (one of the products of EDC synthesis) can cause bubble formation.
