Can Colony PCR be applied to genomic DNA, or only for ligation (vector/insert) screening?

Sure you can. Only thing you might wanna change is the unusual long heating step in the start of a colony PCR which you can reduce in case of genomic DNA.
Colony PCR is also applicable for genomic DNA. This is one of the ways I usually do in screening null mutants and DNA-region integrants.
