I have never observed such kind of inclusion body in A431 cells. In my opinion, the contamination is the most likely. How about the doubling time of the cells?
Dear all, my A431 cells are of course under anti-myco treatment (ciprofloxacin and minocyclin). My DAPI staining after treatments is negative for mycoplasma, whereas definitely positive prior treatments. This is the confirm that I have to carry on myco treatments and perform PCR besides DAPI staining for better detection of contamination levels.
For me it could be just vacuoles, antibiotics can be bit toxic. IMHO DAPI testing is good to confirm infection but not enough to confirm its absence. I read somewhere that detection limit is 100 mycoplasmas per cell.
I agree with you as regards DAPI (as soon as possible I will do a PCR to evaluate treatment efficiency); as regards vacuoles (or of course simple lipid droplets) I not completely agree with you. What strikes me the most is the one inside the nucleus...
