I am looking for Spiroplasma specific conventional PCR primers. I went through some papers but it seems there is no particular primers pair is available as 'Universal'. Can anyone help me sharing their tested primer pair sequence?
Which species of Spiroplasma you want to detect? As far as I know, many of these species are sequenced, so you can design a specific primer based on your interested gene. Otherwise, if you need a universal primer which can detect high variety of spiroplasmas, I recommend you to use primers based on the 16S rDNA gene. For example, you can find some primer in this paper “Spiralin diversity within Iranian strains of Spiroplasma citri”. Although this primer pair design to detect S. citri, I think it can also detect other species of Spiroplasma.
Probably it is impossible to detect spiroplasma from extracted total DNA by 16S rDNA primers, because it can amplified non-specific genome region of mitochondria, or chloroplast. The best is to try for growing in culture medium.
Anyway, if you want to detect S. citri from Total DNA extract I recommend you another primer.
